Indications
- Short-term management of moderate to severe acute post-operative pain
- Relief of pain, inflammation, and light sensitivity after ocular surgery
- Decrease eye inflammation or redness caused by seasonal allergies
Description
- Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Inhibits synthesis of prostaglandins
- Biological activity associated with the S-form
- Linear pharmacokinetic property
- Highly protein bound and largely metabolized in the liver
- Products of metabolism and some unchanged drugs excreted in the urine
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits the cyclooxygenase enzyme system and prostaglandin synthesis
- Demonstrates minimal anti-inflammatory effect at its analgesic dose
Dosage & Administration
- Tablet recommended dose: 10 mg every 4-6 hours
- Injection for acute pain management
- IV bolus or IM administration for postoperative pain
- Different doses for adult and pediatric patients
- Maximum analgesic effect within 1 to 2 hours after dosing
- Duration of analgesic effect usually 4 to 6 hours
- Eye drops dosage: 1 drop in each eye 4 times daily
Interaction
- Increases side effects with other NSAIDs or aspirin
- Enhances anti-coagulant effect with anti-coagulants
- Reduces anti-hypertensive effect with Beta Blockers
- Increases risk of renal impairment with ACE Inhibitors
- Enhances toxicity of methotrexate
Contraindications
- Hypersensitivity to Ketorolac or other NSAIDs
- Avoid use in children under 16 years
- Contraindicated as prophylactic analgesic before surgery

Precautions & Warnings
- Exercise caution in patients over 65 years
- Caution in patients with peptic ulcer, gastrointestinal bleeding, asthma, and liver dysfunction
- Discontinue use in patients with corneal epithelial breakdown and closely monitor corneal health
- Avoid administering while wearing contact lenses
