Indications
- Bronchial asthma
- Bronchospasm
- Chronic obstructive pulmonary disease (COPD)
- Pulmonary disease with spastic bronchial component
Pharmacology
- Doxophylline as a novel bronchodilator
- Selective inhibition of phosphodiesterase 4
- Relaxation of bronchial smooth muscle
- Inhibition of platelet activating factor (PAF)
- Decreased affinities toward adenosine A1 and A2 receptors
Dosage & Administration
- Dosage for elderly: 200 mg tablet two or three times daily
- Dosage for adults: 400 mg tablet two or three times daily
- Dosage for children >12 years of age: 10 ml syrup or 200 mg tablet
- Dosage for children 6-12 years of age: 6-9 mg/kg body weight two times daily
- Special release (SR) tablet for daily dose of 400 mg
Interaction
- Not to be administered with other xanthine preparations
- Limit consumption of caffeine-containing beverages and food
- Exercise caution with ephedrine or other sympathomimetic drugs
- Concurrent administration with erythromycin, TAO, lincomycin, clindamycin, allopurinol, cimetidine, influenza vaccine, propanolol, phenytoin, other anticonvulsants, or cigarette smoking may affect clearance
Contraindications
- Known hypersensitivity to the drug or other xanthine derivatives
- Acute myocardial infarction
- Hypotension
- During lactation

- Insomnia
- Tachycardia
- Extrasystoles
- Tachypnea
- Hyperglycemia
- Albuminuria
- Severe cardiac arrhythmias in case of overdose
Pregnancy & Lactation
- Animal tests show no interference with pre and postnatal growth
- Insufficient clinical evidence regarding effects during pregnancy
- Contraindicated during lactation
- Does not affect the ability to drive or use machines
Precautions & Warnings
- Factors reducing hepatic clearance may increase plasma levels
- Monitoring blood levels recommended in certain cases
- Caution in patients with cardiac disease, hypertension, elderly, hypoxemia, hyperthyroidism, chronic corpulmonale, congestive heart failure, liver disease, peptic ulcer, and renal impairment
- Consider lower dosage in specific cases
Overdose Effects
- Severe cardiac arrhythmias
- Tonic-clonic seizure
- Discontinuation of treatment may be required
- Symptomatic treatment of cardiovascular collapse in case of overdose
