Indications

Mode of Action
- Proton pump inhibitor
- Suppresses gastric acid secretion
- Inhibits H+/K+-ATPase in the gastric parietal cell
- First single optical isomer of proton pump inhibitor
- Better acid control than racemic proton pump inhibitors
- Absorption: Enteric-coated pellet formulation
- Peak plasma levels occur at approximately 1.5 hours
- Distribution: 97% bound to plasma proteins
- Metabolism: Extensively metabolized in the liver by CYP enzyme system
- Excretion: Plasma elimination half-life of approximately 1–1.5 hours
- Combination Therapy with Antimicrobials increases AUC and Cmax
- No clinically significant interactions with certain drugs
Dosage
- Oral dosage form for various conditions
- 20 mg or 40 mg Once Daily for specified periods
- IV Injection or IV infusion for specific patient groups and conditions
Administration
- Tablet or capsule should be swallowed whole and taken one hour before a meal

Interaction
- Extensively metabolized in the liver by CYP2C19 and CYP3A4
- Inhibition of gastric acid secretion may interfere with drug absorption

Contraindications
- Contraindicated in patients with known hypersensitivity to any of the formulation
Side Effects

- Animal studies show no teratogenic effects
- Excretion in milk not studied

- Symptomatic response does not preclude the presence of gastric malignancy

- Use of antacids while taking Esonam
Use in Special Populations
- Safety and effectiveness not established in paediatric patients
- No overall differences in safety and efficacy observed between elderly and younger individuals
- No dosage adjustment recommended for mild to moderate hepatic insufficiency
- Pharmacokinetics not expected to be altered in renal impairment
Overdose Effects
- Acute toxicity signs observed in animal studies
- No reports of overdose with Esonam
- No specific antidote known
- Symptomatic and supportive treatment recommended in case of overdosage
