Indications

Description
- Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2-amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibition of prostaglandin synthesis
- Biological activity associated with the S-form
- Linear pharmacokinetic property
- Highly protein bound
- Metabolized in liver
- Metabolites and unchanged drugs excreted in urine
Pharmacology
- Potent analgesic of non-steroidal anti-inflammatory drugs (NSAIDs)
- Acts by inhibiting the cyclooxygenase enzyme system
- Demonstrates minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet recommended dose: 10 mg every 4-6 hours for short-term use (up to 7 days)
- Injection recommended dose: single or multiple doses for moderately severe acute pain management
- IV bolus administration over at least 15 seconds
- Onset of analgesic effect within 30 minutes
- Maximum analgesic effect in 1 to 2 hours after dosing IV or IM
- Duration of analgesic effect: 4 to 6 hours
- Different dosage recommendations for various age groups and conditions
Interaction
- Increased side effects with other NSAIDs or aspirin
- Enhanced anti-coagulant effect with anti-coagulants
- Reduced anti-hypertensive effect with beta blockers
- Increased risk of renal impairment with ACE inhibitors
- Enhanced methotrexate toxicity
Contraindications
- Hypersensitivity to Ketorolac or other NSAIDs
- Children under 16 years of age
- Prophylactic analgesic use before surgery
