Indications

Description

  • Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
  • Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2-amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibition of prostaglandin synthesis
  • Biological activity associated with the S-form
  • Linear pharmacokinetic property
  • Highly protein bound
  • Metabolized in liver
  • Metabolites and unchanged drugs excreted in urine

Pharmacology

  • Potent analgesic of non-steroidal anti-inflammatory drugs (NSAIDs)
  • Acts by inhibiting the cyclooxygenase enzyme system
  • Demonstrates minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Tablet recommended dose: 10 mg every 4-6 hours for short-term use (up to 7 days)
  • Injection recommended dose: single or multiple doses for moderately severe acute pain management
  • IV bolus administration over at least 15 seconds
  • Onset of analgesic effect within 30 minutes
  • Maximum analgesic effect in 1 to 2 hours after dosing IV or IM
  • Duration of analgesic effect: 4 to 6 hours
  • Different dosage recommendations for various age groups and conditions

Interaction

  • Increased side effects with other NSAIDs or aspirin
  • Enhanced anti-coagulant effect with anti-coagulants
  • Reduced anti-hypertensive effect with beta blockers
  • Increased risk of renal impairment with ACE inhibitors
  • Enhanced methotrexate toxicity

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Children under 16 years of age
  • Prophylactic analgesic use before surgery
