Indications

Pharmacology

  • Inhibitor of gastric acid secretion
  • Blocks hydrogen-potassium-adenosine triphosphatase (H+/K+ ATPase) enzyme system
  • Onset of antisecretory effect within one hour
  • Maximum effect occurring within two hours
  • Inhibition of secretion lasts up to 72 hours
  • Gradual return of secretory activity over 3 to 5 days upon discontinuation

Interaction

Contraindications

  • Known hypersensitivity to omeprazole
  • Need for excluding malignancy before treatment

Side Effects

  • Skin rash, urticaria, and pruritus
  • Photosensitivity, bullous eruption, and erythema multiforme
  • Diarrhoea, headache, constipation, nausea/vomiting, and abdominal pain
  • Dizziness, light headedness, and feeling faint
  • Mental confusion, agitation, depression, and hallucinations
  • Arthritic and myalgic symptoms

Pregnancy & Lactation

  • No adverse effects on pregnancy or the fetus/newborn child based on epidemiological studies
  • Safe use during pregnancy
  • No information on passage into breast milk or effects on the neonate
  • Recommendation to discontinue breast-feeding if essential use is considered

Precautions & Warnings

  • Avoid concomitant use with clopidogrel
  • Association with increased risk for osteoporosis-related fractures
  • Atrophic gastritis noted in long-term treatment
  • Potential for concomitant use with methotrexate leading to toxicities
