Indications
- Treatment of moderate to severe painful conditions

Pharmacology
- Centrally acting synthetic analgesic compound
- Inhibition of reuptake of neurotransmitters serotonin and noradrenaline
- Modification of pain impulse transmission
- Activation of descending serotonergic pathways
- Stimulation of mu-opioid receptors
- Modulation of central monoaminergic inhibitory pathways
Dosage and Administration
- Usual doses of 50 to 100 mg every four to six hours
- Initial dose of 100 mg for acute pain, 50 mg for chronic painful conditions
- Sustained Release Capsule dosage every 12 hours
- Injection dosage of 50-100 mg every 4 to 6 hours
- Suppository dosage of 100 mg Tramadol Hydrochloride 6 hourly
- Total daily dosage by mouth should not exceed 400 mg
- Rectal administration of Tramadol suppository for adults
Interaction
- Monoamine oxidase (MAO) inhibitors not recommended for combination
- Concomitant administration of carbamazepine increases Tendia metabolism
Contraindications
- Hypersensitivity to Tramadol
- Contraindicated in acute intoxication with certain drugs

Pregnancy and Lactation
- Safe use of Tramadol in pregnancy not established
- Tramadol crosses the placenta
- No adequate and well-controlled studies in pregnant women
- Tramadol should not be administered during breastfeeding
Precautions and Warnings
- Respiratory depression risk with large doses
- Not recommended for patients dependent on opioids
- Use with caution when administering with CNS depressants
- Use with great caution in patients taking MAO inhibitors
- Caution in patients with increased intracranial pressure, head injury, acute abdominal conditions
Use in Special Populations
- Tendia can be given in children from the age of 1 year
- Suppository (100 mg Tendia) should not be administered in children and adolescents below the age of 14 years
- Tendia 100 mg SR Capsules not studied in children
