Pharmacology
Triazole antifungal agent, potent inhibitor of fungal cytochrome P-450 dependent enzymes, responsible for the synthesis of ergosterol
Dosage & Administration
- Dosage varies for different indications like Vaginal Candidiasis, Oropharyngeal Candidiasis, Tinea corporis/Tinea cruris/Tinea pedis/Other Tinea, Kerion, Pityriasis versicolor, Onychomycosis, Invasive candidal infections and cryptococcal infections, Prevention of cryptococcal meningitis, Prevention of fungal infections in immunocompromised patients, Superficial Candidiasis, Systemic Candidiasis & Cryptococcal infection
- Specific dosage for children over 1 year based on weight
- Dosage modification for elderly and patients with renal impairment
Interaction
- Increases prothrombin time after warfarin administration
- Prolongs the serum half-life of oral sulphonyl ureas
- Increases plasma concentrations of hydrochlorothiazide
- May increase the level of phenytoin
- Resulted in decreased AUC and shorter half-life when administered with rifampicin
- Calculated interaction with combined oral contraceptive
- Slowly increases cyclosporin concentrations
Contraindications
Known hypersensitivity to Fluconazole or related triazole compounds

Pregnancy & Lactation
Not recommended in pregnancy or in women of childbearing potential unless adequate contraception is employed
Precautions & Warnings
- Abnormalities of hepatic, renal, haematological, and other biochemical function tests observed in some patients
- Rare occurrences of exfoliative cutaneous reactions
- Discontinue therapy on development of rash
- Not recommended during lactation
- Unlikely to impair a patient's ability to drive or use machinery
Use in Special Populations
- Normal dose for the elderly if no evidence of renal impairment
- Dosage adjustments for patients with renal impairment based on creatinine clearance
Overdose Effects
Supportive measures and symptomatic treatment, forced volume diuresis to increase the elimination rate, haemodialysis decreases plasma levels by approximately 50%
