Description
- Sitova is indicated to reduce elevated total cholesterol, LDL cholesterol, apolipoprotein B (Apo-B), and triglycerides levels in patients with various conditions
- Selectively inhibits HMG-CoA reductase, lowering plasma cholesterol and lipoprotein levels
- Rapidly absorbed after oral administration
- Metabolized extensively to ortho- and parahydroxylated derivatives and various beta-oxidation products
- Primarily eliminated in bile following hepatic and/or extra-hepatic metabolism
- Well-tolerated with constipation, flatulence, dyspepsia, and abdominal pain as common side effects
DosageAndAdministration
- Adult dosage range: 10 mg - 80 mg once daily
- Child (10-18 years) dosage range: 10 mg - 20 mg once daily
- Initial dose for adults with familial hypercholesterolaemia: 10 mg, increased to 40 mg once daily and may be further increased to 80 mg once daily
- Started at 10 mg/day for pediatric patients (10-17 years of age) with a maximum recommended dose of 20 mg/day
- Lipid levels should be analyzed within 2 to 4 weeks after initiation
Interaction
- Increased risk of myopathy with concurrent administration of cyclosporine, fibric acid derivatives, niacin, and azole antifungals
- Significant alterations in plasma concentrations when coadministered with antacid, colestipol, digoxin, erythromycin, oral contraceptives, and warfarin
Contraindications
- Hypersensitivity to any component of the medication
- Active liver disease or unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors

- Use with caution in patients who consume substantial quantities of alcohol or have a history of liver disease
- Discontinue therapy if markedly elevated CPK levels occur or myopathy is diagnosed or suspected

- No clinically significant difference in LDL-C reduction between men and women
- No influence on plasma concentrations or LDL-C reduction in renal disease
