Indications
- Reduction of symptoms of intermittent claudication
- Increased walking distance
Pharmacology
- Cilostazol is a quinolinone derivative
- Inhibits cellular phosphodiesterase III (PDE III)
- Suppresses cAMP degradation
- Increases cAMP in platelets and blood vessels
- Inhibits platelet aggregation
- Causes vasodilation
Dosage & Administration
- Recommended dosage: 100 mg bid
- Should be taken at least half an hour before or two hours after meals
- Consider 50 mg bid dosage during coadministration with certain drugs
Interaction
- Omeprazole and Erythromycin increased systemic exposure of Cilosta
- Higher concentrations of Cilosta with concurrent Diltiazem treatment
Contraindications
- Patients with congestive heart failure
- Patients with known or suspected hypersensitivity to any components
Side Effects
- Common: headache, diarrhoea, vomiting, leg cramps, rash
- Less frequent: anorexia, edema
Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Transfer of Cilostazol into milk reported in experimental animals
- Potential risk to nursing infants
Precautions & Warnings
- Use with caution in patients with any degree of heart failure
- No information on concurrent use with Clopidogrel
