Indications
- Reduction of symptoms of intermittent claudication
- Increased walking distance
- Pain, cramps, numbness, or fatigue during exertion
Pharmacology
- Cilostazol as a quinolinone derivative
- Inhibition of cellular phosphodiesterase III (PDE III)
- Suppression of cAMP degradation
- Increase in cAMP in platelets and blood vessels
- Inhibition of platelet aggregation
- Vasodilation
Dosage & Administration
- Recommended dosage: 100 mg bid
- Taken at least 30 minutes before or 2 hours after meals
- Consider 50 mg bid during coadministration of certain drugs
- Pediatric use not established
Interaction
- Omeprazole and Erythromycin increase systemic exposure
- Diltiazem concurrent use leads to higher concentrations
Contraindications
- Contraindicated in patients with congestive heart failure
- Contraindicated in patients with hypersensitivity to components
Side Effects
- Common: headache, diarrhea, vomiting, leg cramps, rash
- Less frequent: anorexia, edema
Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Transfer of Cilostazol into milk reported in animals
- Potential risk to nursing infants
- Discontinue nursing or Cilostazol
Precautions & Warnings
- Use with caution in patients with any degree of heart failure
- No information on concurrent use with Clopidogrel
