Indications
- Peripheral arterial occlusive disease (PAOD) of arteriosclerotic or diabetic origin
- Trophic lesions
- Cerebral vascular diseases
- Circulatory disturbances of the eye
Pharmacology
- Pentoxifylline and its metabolites improve the flow properties of blood
- Increases blood flow to the affected microcirculation
- Enhances tissue oxygenation
- Modifies leukocyte properties
- Increases leukocyte deformability
- Inhibits neutrophil adhesion and activation
Dosage & Administration
- Usual dosage: 400 mg pentoxifylline 2 to 3 times daily
- Tablets are to be swallowed whole during or shortly after a meal with sufficient amounts of liquid
Interaction
- Precautions for use with insulin or oral antidiabetics
- Blood-pressure-lowering effect of antihypertensive agents
- Concomitant administration with theophylline
- Concomitant administration with ciprofloxacin
- Additive effect with platelet aggregation inhibitors
- Concomitant administration with cimetidine
Contraindications
- Hypersensitivity to pentoxifylline or methylxanthines
- Massive bleeding
- Extensive retinal bleeding

- Arrhythmia
- Thrombocytopenia
- Dizziness
- Gastrointestinal disorder
- Pruritus
- Hot flush
- Anaphylactic reaction
Pregnancy & Lactation
- Not recommended during pregnancy
- Passes into breast milk in minute quantities
Precautions & Warnings
- Careful monitoring in patients with severe cardiac arrhythmias
- Particularly careful monitoring in hypotensive patients
- Careful monitoring in patients with impaired renal function
Use in Special Populations
- Dose reduction in hepatic impairment
- Dose reduction in renal impairment
- Treatment must be started at low-dose levels in hypotensive patients
Overdose Effects
- Initial symptoms of acute overdose: nausea, dizziness, tachycardia
- Signs such as fever, agitation, loss of consciousness
Therapeutic Class
- Peripheral Vasodilator drugs: Intermittent Claudication
