Indications

- Sodium Valproate tablet contains Sodium Valproate BP 200 mg
- Sodium Valproate syrup contains Sodium Valproate BP 200 mg
- Sodium Valproate controlled release 200 tablet contains Sodium Valproate BP 133.2 mg & Valproic acid BP 58 mg
- Sodium Valproate controlled release 300 tablet contains Sodium Valproate BP 199.8 mg & Valproic acid BP 87 mg
- Sodium Valproate controlled release 500 tablet contains Sodium Valproate BP 333 mg & Valproic acid BP 145 mg
- Sodium Valproate injection contains Valproic Acid 500 mg
Pharmacology
- Endowed with anti-epileptic activity
- Mechanism may be related to increased brain levels of gamma-aminobutyric acid (GABA)
Dosage & Administration
- Oral administration
- Initial dosage for adults, children (over 20 kg), and children (up to 20 kg)
- Maintenance dose for epilepsy and bipolar disorder
- Injection for adults and children
- Slow intravenous injection over 3-5 minutes
- Continuous or repeated infusion
Interaction
- Acts as a non-specific inhibitor of drug metabolism
- Significant interactions with Phenobarbital, Phenytoin, Warfarin, Aspirin
Contraindications
- Known hypersensitivity to the drug
- Liver dysfunction
- Restricted use during pregnancy and in women of childbearing potential
Side Effects
- Anorexia, nausea, and vomiting
- Effects on the CNS such as sedation, ataxia, and tremor
- Rash, alopecia, and stimulation of appetite
- Elevation of liver enzymes in plasma observed in up to 40% of patients
- Fulminate hepatitis (rare)
- Hepatic injury, acute pancreatitis, and hyperammonemia in children below 2 years of age and those being treated with multiple antiepileptic agents
- Effects on hepatic function
Pregnancy & Lactation
- Exposure to valproate in the first trimester associated with neural tube defects
- Crosses the placenta
- Excreted in breast milk
Precautions & Warnings
- Monitoring of liver functions
- No undue potential for bleeding before starting and before major surgery
- Care in renal impairment, pregnancy, breast-feeding, and systemic lupus erythematosus
- Valporin is partially eliminated in the urine as a ketone metabolite
- Sudden withdrawal of therapy should be avoided
