Pharmacology

  • Novel bronchodilator
  • Inhibition of phosphodiesterase 4
  • Decreased affinities toward adenosine A1 and A2 receptors
  • Inhibition of platelet activating factor (PAF)
  • Inhibition of leukotriene production

Dosage & Administration

  • 200 mg tablet for elderly
  • 400 mg tablet for adults
  • 10 ml syrup for children above 12 years
  • 6-9 mg/kg body weight for children aged 6-12 years
  • Special release (SR) tablet for specific dosages

Interaction

  • Avoid concurrent administration with xanthine preparations
  • Limit consumption of caffeine-containing beverages and food
  • Exercise caution with sympathomimetic drugs
  • Potential interactions with several drugs like erythromycin, TAO, lincomycin, etc.
  • Possible increase in dosage with concurrent use of phenytoin, other anticonvulsants, or cigarette smoking

Contraindications

  • Known hypersensitivity to doxophylline or other xanthine derivatives
  • Acute myocardial infarction

Precautions & Warnings

  • Factors affecting hepatic clearance
  • Monitoring of blood levels in case of influencing factors
  • Caution in patients with cardiac disease, hypertension, elderly, severe hypoxemia, hyperthyroidism, chronic corpulmonale, liver disease, peptic ulcer, renal impairment
  • Special caution in patients with congestive heart failure
  • No risk of addiction or dependence

Overdose Effects
