Indications
- Treatment of panic disorder with or without agoraphobia
- Treatment of Lennox-Gastaut Syndrome (petit mal variant)
- Treatment of akinetic and myoclonic seizures
- Treatment of absence seizures (petit mal)
Pharmacology
- Exhibits anticonvulsive effects
- Exhibits sedative effects
- Exhibits muscle relaxing effects
- Exhibits anxiolytic effects
- Enhances GABAergic neurotransmission
Dosage & Administration
- Initial adult dose for seizure disorders not to exceed 1.5 mg/day
- Initial adult dose for panic disorder is 0.25 mg
- Pediatric dose based on body weight and age
- Injection dosage for infants, children, and adults
Interaction
- Does not alter the pharmacokinetics of phenytoin
- Does not alter the pharmacokinetics of carbamazepine
- Does not alter the pharmacokinetics of phenobarbital
Contraindications
- Patients with history of hypersensitivity to benzodiazepines
- Patients with clinical or biochemical evidence of significant liver disease
- Patients with acute narrow angle glaucoma

- May increase incidence of generalized tonic-clonic seizures
- Concomitant use with valproic acid may produce absence status
Use in Special Populations
- Caution required in pediatric use due to increased bronchial secretion
- Greater pharmacologic effects in geriatric patients
- Renal impairment does not affect pharmacokinetics
- Hepatic impairment affects plasma protein binding
Overdose Effects
- Symptoms include drowsiness, ataxia, and nystagmus
- May lead to areflexia, apnoea, hypotension, and coma
