Indications

• Treatment of endometriosis
• Reduction of pelvic pain in women with endometriosis

Pharmacology

• Acts as an agonist at the progesterone receptor
• Causes endometrial atrophy
• Promotes antiproliferative and antiangiogenic effects
• Reduces endogenous production of oestradiol
• Antagonist at androgen receptors
• Improves androgenic symptoms such as acne and hirsutism

Dosage & Administration

• Dosage of 2 mg daily without any break
• Can be started on any day of the menstrual cycle
• Tablet-taking should be continuous without regard to vaginal bleeding

Interaction

• Metabolized mainly by the cytochrome P450 3A4 system
• Inhibitors of CYP3A4 may affect the progestogen drug metabolism

Contraindications

• Hypersensitivity to dienogest or any excipients
• Active venous thromboembolic disorder
• Arterial and cardiovascular disease
• Diabetes mellitus with vascular involvement
• Severe hepatic disease
• Liver tumors
• Sex hormone-dependent malignancies
• Undiagnosed vaginal bleeding

• Abdominal pain
• Weight increase

Pregnancy & Lactation

• Limited data in pregnant women
• No need to treat endometriosis during pregnancy
• Excretion of Dienogest in breast milk

Precautions & Warnings

• Pregnancy must be excluded before starting treatment
• Use nonhormonal methods of contraception during treatment
• Changes in Bleeding Pattern
• Hepatic Impairment
• Impairment of Fertility

Use in Special Populations

• Not indicated in children prior to menarche
• Safety and efficacy in adolescents not yet established
• No relevant indication for the use in the geriatric population

Overdose Effects

• No specific antidote
• Studies indicate low risk of acute adverse effects
