Indications
- Peripheral arterial occlusive disease (PAOD) of arteriosclerotic or diabetic origin
- Trophic lesions such as leg ulcers and gangrene
- Cerebral vascular diseases
- Circulatory disturbances of the eye in conjunction with degenerative vascular disorders
Pharmacology
- Improves flow properties of blood by decreasing viscosity
- Increases blood flow to affected microcirculation
- Enhances tissue oxygenation
- Produces dose-related hemorrheologic effects
- Lowers blood viscosity
- Improves erythrocyte flexibility
- Modifies leukocyte properties
- Increases leukocyte deformability
- Inhibits neutrophil adhesion and activation
- Increases tissue oxygen levels
Dosage & Administration
- Usual dosage: 400 mg pentoxifylline 2 to 3 times daily
- Tablets are to be swallowed whole during or shortly after a meal with sufficient amounts of liquid
Interaction
- Potentiation of blood-sugar-lowering effect of insulin or oral antidiabetics
- Increased anti-coagulant activity with concomitant use of Vasolax and anti-vitamin K
- Increased blood-pressure-lowering effect with antihypertensive agents
- Increased theophylline levels with concomitant administration of Vasolax and theophylline
- Increased serum concentration of Vasolax with concomitant administration with ciprofloxacin
- Additive effect with platelet aggregation inhibitors
Contraindications
- Hypersensitivity to pentoxifylline, other methylxanthines, or any excipients of Pentoxifylline
- Massive bleeding
- Extensive retinal bleeding
Side Effects
- Transaminases increased
- Blood pressure decreased
- Arrhythmia
- Tachycardia
- Thrombocytopenia
- Leucopenia/neutropenia
- Dizziness
- Headache
- Pruritus
- Erythema
- Urticaria
- Rash
- Gastrointestinal disorders such as nausea, vomiting, diarrhea, and constipation
- Vascular disorders like hot flush and hemorrhage
- Anaphylactic reactions
- Cholestasis
- Agitation
- Sleep disorders
Pregnancy & Lactation
- Not recommended during pregnancy due to insufficient experience
- Physician must carefully weigh possible risks and benefits before administering in breast-feeding women
Precautions & Warnings
- Discontinue Vasolax at first signs of an anaphylactic/anaphylactoid reaction
- Careful monitoring in patients with severe cardiac arrhythmias
- Hypotensive patients
- Patients with impaired renal and liver function
- Patients treated concomitantly with antidiabetic agents, ciprofloxacin, theophylline, anti-vitamin K, or platelet aggregation inhibitors
Use in Special Populations
- Dose reduction in patients with severely impaired liver function
- Dose reduction in patients with impairment of renal function
- Low-dose start in hypotensive patients or patients at risk from a reduction in blood pressure
Overdose Effects
- Initial symptoms may include nausea, dizziness, tachycardia, or a fall in blood pressure
- Signs like fever, agitation, flush, loss of consciousness, areflexia, tonic-clonic convulsions, and coffee-ground vomiting may occur
Therapeutic Class
- Peripheral Vasodilator drugs for Intermittent Claudication
Storage Conditions
- Keep in a cool and dry place, away from light
- Do not use later than date of expiry
- Keep out of the reach of children
- To be dispensed only on the prescription of a registered physician
