Indications
- Treatment of acute and chronic psychoses
- Treatment of mania
- Treatment of schizophrenia
Pharmacology
- Selective monoaminergic antagonist
- High affinity for serotoninergic 5-HT2 and dopaminergic D2 receptors
- Binds to alpha1 adrenergic receptors
- Binds with lower affinity to H1 histamine and alpha2 adrenergic receptors
- Potent D2 antagonist
Dosage & Administration
- Initial dose of 2 mg in 1-2 divided doses on 1st day
- Usual dose range of 4-6 mg daily
- Maximum dose of 16 mg daily
- Dosing adjustments for elderly or patients with hepatic or renal impairment
- Different dosage recommendations for psychoses, mania, and schizophrenia
Interaction
- Antagonizes the effects of levodopa and dopamine agonists
- Reduces plasma clearance when administered with Carbamazepine
- May decrease clearance when administered with Clozapine
- Enhances the effects of certain antihypertensives
