Pharmacology

  • Active form of Vitamin D3: Calcitriol
  • Vitamin D receptors (VDRs) binding
  • Fat-soluble with a half-life of about 50 days
  • Absorption in the small intestine
  • Metabolism to 25-hydroxy Vitamin D3 (Calcidiol)
  • Second hydroxylation to 1,25-dihydroxy Vitamin D3 (Calcitriol)
  • Absorption of calcium
  • Storage in tissues such as fat and muscle
  • Elimination via faeces and urine

Contraindications

  • Overdose in infants induced by nursing mothers not observed
  • Consideration of dose for additional vitamin D3 given to the mother
