Description

Indication
- Reduction of total cholesterol and LDL cholesterol in patients with heterozygous and homozygous familial hypercholesterolaemia
- Treatment of patients with elevated serum triglyceride levels in hypertriglyceridaemia
- Reduction of cardiac ischaemic events in patients with asymptomatic or mild to moderate symptomatic coronary artery disease
- Reduction of cholesterol and triglycerides in patients with mixed dyslipidemia
- Treatment of patients with dysbetalipoproteinaemia
- Reduction of cholesterol concentrations in patients with hypercholesterolemia associated with diabetes mellitus or renal transplantation
Dosage administration
- Adults: Usually 10 mg once daily, may be increased to max. 80 mg once daily if necessary
- Child (10-18 years): Initially 10 mg once daily, increased if necessary to max. 20 mg once daily
- Dosing range: 10 to 80 mg once daily
- Can be administered as a single dose at any time of the day, with or without food
Interactions
- Concurrent administration with cyclosporine, fibric acid derivatives, niacin, erythromycin, azole antifungals increases risk of myopathy
- Plasma concentrations of atorvastatin decreased when coadministered with antacids or colestipol and increased when coadministered with erythromycin
- Increased plasma digoxin concentrations when coadministered with digoxin
- Coadministration with oral contraceptives and warfarin requires consideration of increased AUC values and prothrombin time, respectively
Contraindications
- Hypersensitivity to any component of Frenvas
- Active liver disease or unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors

Precautions warnings

- Discontinue therapy if markedly elevated CPK levels occur or myopathy is diagnosed or suspected
- Use with caution in patients with substantial alcohol consumption or history of liver disease
Special populations
- Higher plasma concentrations in healthy elderly subjects
- Greater degree of LDL-lowering in the elderly patient population
- No clinically significant difference in LDL-C reduction between men and women
- Renal disease has no influence on plasma concentrations or LDL-C reduction
- Markedly increased plasma concentrations in patients with chronic alcoholic liver disease

- Specific treatment not available for atorvastatin overdose
- Symptomatic and supportive measures instituted
- Serum CK levels should be monitored
