Indications
- Seasonal Allergic Rhinitis
- Perennial Allergic Rhinitis
- Urticaria
Pharmacology
- Long-acting
- Non-sedative
- Histamine H1-receptor antagonist
- Platelet activating factor (PAF) antagonist
- Better therapeutic effect than isolated antihistamine
- Antiallergic properties such as inhibition of mast cell degranulation and cytokine release
Dosage & Administration
- Recommended dose for adults and adolescents: 10 mg once a day
- Recommended dose for children aged 2 to 11 years based on weight
- May be taken with or without food
Interaction
- Increased systemic exposure with ketoconazole or erythromycin
- Caution with CYP3A4 inhibitors, statins, CNS depressants, and alcohol
- Avoid concomitant use with grapefruit and grapefruit juice
Contraindications
- Hypersensitivity to Rupatadine or excipients
Side Effects
- Common: Asthenia, dizziness, drowsiness
- Uncommon: Appetite increased, arthralgia, back pain, gastrointestinal discomfort, nausea, rash, weight increased
- Rare: Palpitations, tachycardia
Pregnancy & Lactation
- Avoid use during pregnancy unless potential benefit outweighs potential risk
- Not recommended during lactation unless benefits justify the risk
Precautions & Warnings
- Use with caution in patients with prolonged QT interval, uncorrected hypokalemia, ongoing proarrhythmic conditions, or elderly patients
- Not recommended in patients with impaired kidney or liver function
Use in Special Populations
- Use with caution in elderly patients
- Not established for patients less than 12 years of age
- Not recommended in patients with renal or hepatic insufficiency
Overdose Effects
- Most common adverse reaction is somnolence
- Symptomatic treatment and supportive measures for accidental ingestion of very high doses
