Description

Indications
- Acute and delayed nausea and vomiting
- Uncontrolled nausea and vomiting
- Chemotherapy-induced nausea and vomiting (CINV)
- Radiotherapy-induced nausea and vomiting (RINV)
- Post-operative & Post-discharge nausea and vomiting (PONV & PDNV)
Mode of Action
- Palonosetron is a 5-HT3 receptor antagonist with strong binding affinity for this receptor and little or no affinity for other receptors. It works by blocking the actions of Serotonin at 5-HT3 receptors. It is likely that Palonosetron works in the small intestine but it may also work in the brain. Pharmacokinetics include linear dose-proportional behavior, volume of distribution, metabolism percentages, excretion, total body clearance, renal clearance, and longer half-life.
Dosage & Administration
- Usual dosage for adults, chemotherapy-induced nausea and vomiting dosage, radiotherapy-induced nausea and vomiting dosage, post-operative nausea and vomiting dosage, and children dosage.
Interaction
- Safely administered with corticosteroids, analgesics, antiemetics/antinauseants, antispasmodics, and anticholinergic agents. No inhibition of the antitumor activity of certain drugs. No significant pharmacokinetic interactions with metoclopramide. Not an inhibitor or inducer of specific enzymes.
Contraindications
- Contraindicated in patients known to have hypersensitivity to the drug or any of its components.
Side Effects
- The most common adverse reactions are headaches and constipation.
Pregnancy & Lactation
- Pregnancy category 'B'. It is not known whether Palonosetron is excreted in breast milk.
Use in Special Populations
- No dosage adjustment is recommended in elderly patients >65 years of age. Use in children (1 month to 10 years) with specific dosage. No dosage adjustment is recommended in patients with renal and hepatic dysfunction.
Overdose Effects
- There is no known antidote to Vomiren. Overdose should be managed with supportive care.
