Indications
- Treatment of all forms of dementia of the Alzheimer's type
- May be indicated in other types of dementia
Pharmacology
- Low to moderate affinity as an uncompetitive (open-channel) NMDA receptor antagonist
- Well absorbed after oral administration
- Excreted predominantly unchanged in the urine
- Terminal elimination half-life of about 60-80 hours
- Linear pharmacokinetics over the therapeutic dose range
Dosage & Administration
- Recommended maintenance dose for adults and older patients: 20 mg every day
- Upward titration with 5 mg per week over 3 weeks to achieve maintenance dose
- Different dosage guidelines for renal and hepatic impairment
- Not recommended for use in children below 18 years
Interaction
- Enhancement of effects of L-dopa, dopaminergic agonists, and anticholinergics
- Reduction in effects of barbiturates and neuroleptics
- Contraindicated with specific medications such as amantadine, ketamine, and phenytoin
Contraindications
- Hypersensitivity to Memantine Hydrochloride or to any excipients used in the formulation
