Pharmacology
- Absorption: Rapidly absorbed after oral administration, with maximum plasma concentrations occurring within 1 to 2 hours
- Distribution: 98% bound to plasma proteins, extensive distribution in the body
- Metabolism: Extensively metabolized to active derivatives, primarily metabolized by cytochrome P450 3A4
- Excretion: Eliminated primarily in bile, mean plasma elimination half-life is approximately 14 hours in humans
Interaction
- Antacid: Coadministration may decrease plasma concentrations of atorvastatin
- Colestipol: Coadministration may decrease plasma concentrations but increase LDL-C reduction
- Digoxin: Plasma digoxin concentrations may increase by approximately 20% with coadministration
- Erythromycin: Plasma concentrations of atorvastatin may increase approximately 40% with coadministration
Contraindications
Should not be used in patients with hypersensitivity to any component, active liver disease, unexplained elevations of serum transaminases, or history of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors
