Description

  • Orally active cephalosporin antibiotic with bactericidal activity
  • Effective against a wide variety of Gram-positive and Gram-negative organisms

Indications

  • Treatment for Upper Respiratory Tract Infections
  • Treatment for Urinary Tract Infections
  • Clinical efficacy against various pathogens
  • High stability in the presence of beta-lactamase enzymes

Pharmacology

  • Third generation semisynthetic cephalosporin antibiotic
  • Bactericidal against a broad spectrum of bacteria
  • Kills bacteria by interfering in the synthesis of bacterial cell wall
  • Highly stable in the presence of Beta-lactamase enzyme
  • Absorption around 40% to 50% with or without food

Dosage & Administration

  • Usual course of treatment is 7 days, extendable to 14 days if required
  • Recommended adult dosage is 200-400 mg daily according to infection severity
  • Dosage recommendations for elderly, children, and in renal impairment cases

Interaction

  • Potential increases in prothrombin times in some patients receiving anticoagulant therapy

Contraindications

  • Hypersensitivity to cephalosporin antibiotics

Side Effects

  • Majority of adverse reactions observed in clinical trials were mild and self-limiting
  • Common gastrointestinal disturbances: diarrhea, nausea, abdominal pain
  • Other side effects such as headache, dizziness, hypersensitivity reactions, hematological and clinical chemistry changes
  • Rare occurrences of pseudomembranous colitis and mild transient changes in liver and renal function tests

Pregnancy & Lactation

  • No adequate and well-controlled studies in pregnant women
  • Caution advised when administered to nursing women

Precautions & Warnings

  • Caution in patients with known hypersensitivity to other drugs
  • Consideration for cross-allergenicity between penicillins and cephalosporins
  • Administration with caution in patients with markedly impaired renal function
  • Alteration of normal flora of the colon leading to antibiotic-associated diarrhea
  • Recommendation against use in pregnancy or nursing mothers unless considered essential by the physician

Overdose Effects

  • Gastric Lavage may be indicated in case of overdose
  • No specific antidote available and not significantly removed by dialysis
  • Adverse reactions in healthy volunteers receiving single doses
