Indications
- Reduction of myocardial infarction (MI)
- Reduction of stroke in patients with non-ST-segment elevation ACS
- Reduction of MI and stroke in patients with acute ST-elevation MI
- Reduction of MI and stroke in patients with established peripheral arterial disease or recent MI/stroke
Pharmacology
Inhibition of platelet activation and aggregation
Dosage & Administration
- 300 mg oral loading dose for acute coronary syndrome
- 75 mg once daily orally for recent MI, recent stroke, or established peripheral arterial disease
Interaction
- Increased bleeding risk with NSAIDs, warfarin, SSRIs, and SNRIs
- Avoid concomitant use with CYP2C19 inhibitors and Repaglinide
Contraindications
- Hypersensitivity to the drug substance or any component
- Active pathological bleeding
Side Effects
- Common: Bleeding, diarrhea, gastrointestinal discomfort, haemorrhage, skin reactions
- Rare: Acquired haemophilia, anaemia, angioedema, arthralgia, bone marrow disorders
Pregnancy & Lactation
- Use only if clearly needed during pregnancy
- Unknown excretion in human breast milk
Precautions & Warnings
- Impaired metabolism by genetic variations and CYP2C19 inhibitors
- Increased risk of bleeding
- Risk of cardiovascular events upon discontinuation
- Potential for Thrombotic Thrombocytopenic Purpura (TTP)
- Hypersensitivity reactions
