Description
- Algaba is indicated for neuropathic pain, postherpetic neuralgia, partial-onset seizures, fibromyalgia, and neuropathic pain associated with spinal cord injury.
- Pregabalin is a structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA) and has high affinity to the alpha2-delta site in the central nervous system.
- Dosage varies based on indications, patient's age, and creatinine clearance level. Maximum recommended daily doses for different indications are provided. Pregabalin CR capsule dose conversion from pregabalin tablets is also mentioned.
- Algaba is taken orally and can be taken with or without food. Pregabalin CR tablet should be administered after an evening meal, swallowed whole, and should not be split, crushed, or chewed.
- Algaba is unlikely to be involved in significant pharmacokinetic drug interactions. There are no significant drug interactions with food or others.
- Algaba is contraindicated in patients with known hypersensitivity to Pregabalin or any of its components. Not recommended for use during pregnancy and lactation.
- Precautions and warnings include the possibility of angioedema, increased risk of suicidal thoughts or behavior, potential for respiratory depression, and caution in coadministration with thiazolidinedione antidiabetic agents.
- Use in special populations is not established for pediatric patients, and it is not recommended during pregnancy and lactation.
- In case of overdose, reduced consciousness, depression/anxiety, confusional state, agitation, and restlessness can occur. General supportive care and observation of the clinical status of the patient are indicated.
- Pharmacological class: Adjunct anti-epileptic drugs, Primary anti-epileptic drugs.
- Storage: Keep in a cool & dry place, protected from light & moisture, and out of the reach of children.
Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology

Dosage
- Neuropathic pain associated with diabetic peripheral neuropathy in adults (DPN): 50-300 mg/day based on efficacy and tolerability
- Postherpetic neuralgia in adults (PHN): 75-300 mg/day based on efficacy and tolerability
- Management of fibromyalgia in adults: 300-450 mg/day based on efficacy and tolerability
- Neuropathic pain associated with spinal cord injury in adults: 150-600 mg/day based on efficacy and tolerability
Administration
- Route of administration: Oral, with or without food
- Pregabalin CR tablet administration: After an evening meal, swallow whole, do not split, crush, or chew

Precautions warnings
- Angioedema
- Discontinue immediately in case of hypersensitivity reactions
- Increased risk of suicidal thoughts or behavior
- Respiratory depression possible with concomitant CNS depressants or underlying respiratory impairment
- May cause dizziness and somnolence, caution with driving or operating machinery
- Withdraw gradually over a minimum of 1 week to avoid increased seizure frequency or adverse reactions
- May cause peripheral edema, caution with coadministration with thiazolidinedione antidiabetic agents

- Use in children and adolescents: Safety and effectiveness not established for specific indications and age groups
