Indications
- Relief of signs and symptoms of osteoarthritis
- Relief of signs and symptoms of rheumatoid arthritis
Pharmacology
- Non-steroidal anti-inflammatory drug (NSAID)
- Exhibits anti-inflammatory properties
- Exhibits analgesic properties
- Exhibits antipyretic properties
- Mode of action is not fully known
- Inhibits prostaglandin synthesis
Dosage & Administration
- Available in 500 mg & 750 mg tablets
- Also available in 1000 mg tablet
- Oral administration
- Recommended starting dose is 1000 mg
- Can be taken with or without food
- Highest studied dose is 2000 mg per day
- Dosage should be adjusted for renal insufficiency
- Not studied in pediatric patients
Interaction
- May diminish antihypertensive effect of ACE-inhibitors
Contraindications
- Hypersensitivity to nabumetone or its excipients
- Asthma, urticaria, or allergic-type reactions after taking aspirin or other NSAIDs
- Severe, rarely fatal, anaphylactic-like reactions to NSAIDs
- Contraindicated for peri-operative pain in the setting of coronary artery bypass graft (CABG) surgery
Side Effects
- Gastrointestinal: diarrhea, dyspepsia, abdominal pain, constipation, flatulence, nausea, positive stool guaiac, dry mouth, gastritis, stomatitis, vomiting
- Central Nervous System: dizziness, headache, fatigue, increased sweating, insomnia, nervousness, somnolence
- Dermatologic: pruritus, rash
- Special Senses: tinnitus
- Miscellaneous: edema
Pregnancy & Lactation
- Pregnancy Category C
- Not recommended during third trimester of pregnancy
- Not recommended for use in nursing mothers
Precautions & Warnings
- Associated with renal papillary necrosis and abnormal renal pathology in animals
- May cause reduction in renal blood flow
- Patients at greatest risk are those with impaired renal function, heart failure, liver dysfunction, those taking diuretics, and the elderly
- Discontinuation may lead to recovery
Overdose Effects
- Symptoms may include lethargy, drowsiness, nausea, vomiting, epigastric pain
- Rare occurrences of hypertension, acute renal failure, respiratory depression, and coma
- Management involves symptomatic and supportive care
- No specific antidotes available
