Uses and Administration
- Monotherapy in the treatment of partial-onset seizures in adults and adolescents from 16 years of age
- Adjunctive therapy in the treatment of partial-onset seizures in adults, adolescents, children, and infants from 1 month of age with epilepsy
- Treatment of myoclonic seizures in adults and adolescents from 12 years of age with Juvenile Myoclonic Epilepsy
- Treatment of primary generalized tonic-clonic seizures in adults and adolescents from 12 years of age with Idiopathic Generalised Epilepsy
- Oral administration in two equally divided doses daily
- Preferred oral solution for use in infants and children under the age of 6 years
Dosage and Administration
- Starting dose of 250 mg twice daily for adults and adolescents from 16 years of age
- Initial therapeutic dose of 500 mg twice daily for adults and adolescents (≥18 years) weighing 50 kg or more
- Incremental dosage adjustments based on clinical response
- Maximum dose of 1500 mg twice daily for adults and adolescents
- Tablet formulation not adapted for use in infants and children under the age of 6 years
Interaction with Other Medications
- Minimal influence on serum concentrations of existing antiepileptic medicinal products
- No influence on the pharmacokinetics of antiepileptic medicinal products
- Potential interaction with probenecid, methotrexate, and laxatives
- No alteration of Levepro absorption by food, but a slightly reduced rate
Contraindications
- Hypersensitivity to the active substance or other pyrrolidone derivatives or to any of the excipients
Side Effects
- Common adverse reactions include somnolence, asthenia, infection, dizziness in adult patients
- Common adverse reactions include fatigue, aggression, nasal congestion, decreased appetite, irritability in pediatric patients
Pregnancy and Lactation
- Not recommended during pregnancy and in women of childbearing potential not using contraception unless clinically necessary
- Excreted in human breast milk, therefore breastfeeding not recommended
Precautions and Warnings
- Dose adjustment may be required for patients with renal impairment
- Rare cases of decreased blood cell counts have been described
- Possible association with suicide, suicidal ideation, and behavior
- Long-term effects on growth, puberty, and childbearing potential in children remain unknown
Use in Special Populations
- Dose adjustment recommended in elderly patients with compromised renal function
- Dose individualization according to renal function
- No dose adjustment needed in patients with mild to moderate hepatic impairment
Overdose Effects and Treatment
- Occurrence of somnolence, agitation, aggression, depressed level of consciousness, respiratory depression, and coma
- No specific antidote available
- Symptomatic treatment may include haemodialysis
- Extraction efficiency of Levepro on dialysis is 60%
