Indications
- Treatment of thiamine deficiency manifestations
- Treatment of Beriberi and Wernick's encephalopathy
- Treatment of neuritis associated with pregnancy
- Treatment of pellagra
- Prophylactic use in low dietary intake or impaired absorption conditions
- Prophylactic use in pregnancy and carbohydrate-rich diet
Pharmacology
- Coenzyme for decarboxylation of α-ketoglutaric acid
- Affects peripheral nervous system, gastrointestinal tract, cardiovascular system
- Necessary for optimal growth of infants and children
- Not stored in the body, regularly lost during deficiency
- Rapid absorption and distribution throughout the body
- Excess ingested thiamine eliminated in urine
- Plasma half-life of 24 hours
Dosage & Administration
- Prophylaxis: 3 to 10 mg daily
- Mild chronic deficiency: 10 to 25 mg daily
- Severe deficiency: 200 to 300 mg daily

- No hazardous drug interactions reported
- Acts synergistically with other B-complex vitamins
- Potential for adverse effects considerably reduced
Contraindications
- No absolute contraindication
- Increased risk of anaphylaxis with repeated parenteral administration
- Mild allergic phenomena as warning signs
- Avoid intravenous administration, especially in comatose patients
- No need for parenteral administration once deficiency corrected

- Anaphylactic reactions with large intravenous doses in sensitive patients
