Indications
- Treats hypertension
- Lowers blood pressure

Pharmacology
- Contains Levamlodipine
- Indicated for adults and pediatric patients 6 years and older
- Inhibits transmembrane influx of calcium ions
- Selective effect on vascular smooth muscle cells
- Bioavailability of 64-90%
- Volume of distribution similar to amlodipine
- Metabolized to inactive metabolites
- Metabolism mediated by CYP3A4
- 60% eliminated in urine
- Tmax of 6-12 h
Dosage & Administration
- Oral route of administration
- Usual initial dose of 2.5 mg once daily
- Maximum dose of 5 mg once daily
- Lower initial dose for elderly and hepatically impaired patients
- Adjust dosage according to blood pressure goals
- Effective pediatric dose for ages 6-17 years is 1.25-2.5 mg once daily
Interaction
- CYP3A Inhibitors increase systemic exposure
- Impact on simvastatin and immunosuppressants
- Can be taken with or without food
- Avoid grapefruit products & natural licorice
Contraindications
- Contraindicated in patients with known sensitivity to amlodipine
Side Effects
- Common: Difficulty in breathing, Dizziness, Reflex Tachycardia, Hypotension, Abdominal Pain & Edema
- Rare: Diarrhea, Back pain, Headache, Fatigue & Nausea
Pregnancy & Lactation
- Limited data on use in pregnant women
- No adverse effects on breastfed infants observed
Precautions & Warnings
- Possible symptomatic hypotension
- Worsening angina and acute myocardial infarction
- Titrate slowly for patients with severe hepatic impairment
Overdose Effects
- Expect excessive peripheral vasodilation
- Marked hypotension
- Possibly reflex tachycardia
