Medicine Details

Indications

  • Treatment of type 2 diabetes mellitus in adults
  • Treatment of type 2 diabetes mellitus in children from 10 years of age and adolescents
  • Reduction of diabetic complications in overweight type 2 diabetic adult patients

Pharmacology

  • Lowering of basal and postprandial plasma glucose
  • Decrease in hepatic glucose production
  • Improvement in insulin sensitivity
  • Increase in peripheral glucose uptake and utilization

Dosage & Administration

  • Usual starting dose of Metformin immediate release tablet for adults: 500mg twice a day or 850mg once a day
  • Maximum daily dose of Metformin immediate release tablet for adults: 2550mg
  • Usual starting dose of Metformin extended release tablet for adults: 500mg once daily with the evening meal
  • Usual starting dose of Metformin immediate release tablet for children: 500mg twice a day
  • Maximum daily dose of Metformin immediate release tablet for children: 2000mg

Interaction

Contraindications

  • Hypersensitivity to the active substance or to any of the excipients
  • Acute metabolic acidosis
  • Severe renal failure
  • Conditions with the potential to alter renal function
  • Conditions which may cause tissue hypoxia

Side Effects

  • Hemolytic anemia
  • Lactic acidosis
  • Decrease of vitamin B12 absorption
  • Peripheral neuropathy in patients with vitamin B12 deficiency
  • Taste disturbance
  • Encephalopathy
  • Gastrointestinal disorders
  • Liver function test abnormalities or hepatitis
  • Skin reactions

Pregnancy & Lactation

  • Uncontrolled diabetes during pregnancy associated with increased risk of congenital abnormalities and perinatal mortality
  • Metformin excreted into human breast milk
  • Breastfeeding not recommended during metformin treatment

Precautions & Warnings

  • Substantially excreted by the kidney
  • Lowering of vitamin B12 level
  • Increased risk of hypoglycemia
  • Regular monitoring of renal function advised

Use in Special Populations

  • Elderly subjects may require adjusted metformin dosage based on renal function
  • Careful follow-up of the effect of metformin on growth and puberty in children

Overdose Effects
