Indications

• Prevention and treatment of secondary hyperparathyroidism associated with Chronic Kidney Disease (CKD) Stages 3 and 4
• Prevention and treatment of secondary hyperparathyroidism associated with CKD Stage 5 in patients on hemodialysis (HD) or peritoneal dialysis (PD)

Pharmacology

• Paricalcitol is a synthetic, biologically active vitamin D2 analog of calcitriol
• Biological actions are mediated through binding of the VDR, resulting in the selective activation of vitamin D responsive pathways
• Reduction of parathyroid hormone levels by inhibiting PTH synthesis and secretion

Dosage & Administration

• May be taken without regard to food
• Recommendations for administration in Chronic Kidney Disease (CKD) Stages 3 and 4 in Adults
• Recommendations for administration in Chronic Kidney Disease Stage 5 in Adults

Interaction

• Interaction with strong CYP3A inhibitors
• Reduced absorption if administered simultaneously with cholestyramine or mineral oil

Contraindications

• Patients with evidence of hypercalcemia or vitamin D toxicity

Side Effects

• Adverse reaction rates observed in clinical studies

Pregnancy & Lactation

• Limited data with Paricalcitol capsules in pregnant women
• No information available on the presence of paricalcitol in human milk
