Indications

  • Treatment of partial-onset seizures in adults and adolescents
  • Adjunctive therapy for myoclonic seizures in adults and adolescents
  • Treatment of primary generalized tonic-clonic seizures in adults and adolescents

Description

Mechanism of action is unknown, inhibits burst firing without affecting normal neuronal excitability

Dosage

  • Adults and adolescents from 16 years of age: Recommended starting dose is 250 mg twice daily, can be increased to a maximum of 1500 mg twice daily
  • Pediatric population: Not adapted for use in infants and children under the age of 6 years, oral solution preferred for this population

Administration

Tablets must be taken orally, swallowed with liquid, administered in two equally divided doses

Interaction

  • Leviceta did not influence the serum concentrations of existing antiepileptic medicinal products
  • Probenecid inhibits the renal clearance of the primary metabolite
  • Concomitant administration of Leviceta and methotrexate has been reported to decrease methotrexate clearance

Contraindications

Hypersensitivity to the active substance or other pyrrolidone derivatives or to any of the excipients

Pregnancy & Lactation

  • Not recommended during pregnancy and breastfeeding

Precautions & Warnings

  • Renal impairment may require dose adjustment
  • Use has been associated with acute kidney injury
  • Rare cases of decreased blood cell counts
  • Suicidal ideation and behavior reported in patients treated with anti-epileptic agents

Use in Special Populations

  • Elderly patients may need dose adjustment
  • Renal function must be considered for dose individualization
  • No dose adjustment needed in patients with mild to moderate hepatic impairment

Overdose Effects

  • Somnolence
  • Agitation
  • Aggression
  • Depressed level of consciousness
  • Respiratory depression
  • Coma observed with Leviceta overdoses
  • No specific antidote available
  • Haemodialysis may be included in the treatment of an overdose
