Indications
- Monotherapy for partial-onset seizures in adults

Description
- Precise mechanism of action unknown
- Minimal activity in submaximal stimulation
- Protection against secondarily generalized activity from focal seizures
- Inhibition of burst firing without affecting normal neuronal excitability
- Prevention of hyper synchronization of epileptiform burst firing
Dosage
- Recommended starting dose for adults and adolescents from 16 years of age: 250 mg twice daily
- Initial therapeutic dose for adults and adolescents (≥18 years) and adolescents (12 to 17 years) weighing 50 kg or more: 500 mg twice daily
- Maximum dose: 1500 mg twice daily
- Tablet formulation not adapted for use in infants and children under 6 years
- Oral solution preferred for pediatric population
Administration
- Tablets to be taken orally with sufficient liquid
- Daily dose administered in two equally divided doses
Interaction
- No influence on serum concentrations of existing antiepileptic medicinal products
- Inhibition of renal clearance of the primary metabolite by probenecid
- Decrease in methotrexate clearance when administered concomitantly with Levefix
- Isolated reports of decreased Levefix efficacy when concomitantly administered with oral macrogol laxative
Contraindications
- Hypersensitivity to the active substance or other pyrrolidone derivatives or to any of the excipients
Side Effects
- Most common adverse reactions: somnolence, asthenia, infection, dizziness in adult patients
- Most common adverse reactions: fatigue, aggression, nasal congestion, decreased appetite, irritability in pediatric patients
Pregnancy & Lactation
- Not recommended during pregnancy and in women of childbearing potential not using contraception
- Excreted in human breast milk, breastfeeding not recommended
Precautions & Warnings
- Dose adjustment required for patients with renal impairment
- Very rarely associated with acute kidney injury
- Rare cases of decreased blood cell counts described at the beginning of treatment
- Increased risk of suicidal thoughts and behavior reported in patients treated with anti-epileptic agents
- Tablet formulation not adapted for use in infants and children under 6 years
Use in Special Populations
- Dose adjustment recommended in elderly patients with compromised renal function
- Daily dose must be individualized according to renal function
Overdose Effects
- Observation of somnolence, agitation, aggression, depressed level of consciousness, respiratory depression, and coma
- No specific antidote
- Symptomatic treatment may include haemodialysis
