Indications
- Neuropathic Pain
- Adjunctive therapy in partial seizure and secondary generalized seizure
Pharmacology
- Gabapentin is an anti-convulsant
- Structural analog of gamma-amino-butyric-acid (GABA)
- Binds with the alpha-2-delta subunit of voltage gated L-type Calcium channel
- Inhibits branched chain amino acid transferase
- Inhibits neurotransmitter release of excitatory amino acid
Dosage
- Initiate as a single 300 mg dose on Day-1
- 300 mg twice on Day-2
- 300 mg thrice on Day-3
- Titrate up as needed for pain relief to a daily dose of 1800 mg (divided TID)
- Effective dose of 900 to 1800 mg/day given in three divided doses for patients over 12 years of age
- Starting dose of 300 mg three times a day for patients over 12 years of age
- Starting dose should range from 10-15 mg/kg/day in 3 divided doses for pediatric patients age 3-12 years
- Effective dose reached by upward titration over a period of approximately three days for pediatric patients age 3-12 years
Administration
- Oral administration
- Can be taken with or without food
Interaction
- Antacids may reduce the bioavailability of GBR by up to 20%
- Cimetidine may alter its renal excretion
- Does not interact with other anti-epileptic drugs or with oral contraceptive preparations
Contraindications
- Known hypersensitivity to the drug

Pregnancy & Lactation
- Pregnancy category C
- Use during pregnancy only if potential benefits justify potential risk to the fetus

Precautions & Warnings
- Take GBR only as prescribed
- Avoid driving or operating machinery until effects on mental and/or motor performance are known
Use in Special Populations
- Dose reduction in renal impaired patients
