Indications

  • Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
  • Postherpetic neuralgia (PHN)
  • Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
  • Fibromyalgia
  • Neuropathic pain associated with spinal cord injury

Pharmacology

  • Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
  • Binds with high affinity to the alpha2-delta site in central nervous system tissues

Dosage

  • Maximum recommended dose of Pregabalin for neuropathic pain associated with diabetic peripheral neuropathy in adults
  • Recommended dose of Pregabalin for postherpetic neuralgia in adults
  • Recommended dose of Pregabalin for management of fibromyalgia in adults
  • Recommended dose range of Pregabalin for neuropathic pain associated with spinal cord injury in adults

Administration

  • Route of administration: Oral
  • Pregabalin CR tablet administration: After an evening meal, swallow whole, not split, crushed or chewed

Drug interaction

  • Unlikely to be involved in significant pharmacokinetic drug interactions

Contraindications

  • Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components

  • Edema
  • Blurred vision
  • Weight gain
  • Thinking abnormal
  • Increased weight (in pediatric patients for the treatment of partial onset seizures)
  • Increased appetite (in pediatric patients for the treatment of partial onset seizures)

Pregnancy & Lactation

  • No adequate and well-controlled studies with pregabalin in pregnant women
  • Small amounts of pregabalin detected in the milk of lactating women
  • Breastfeeding not recommended

Precautions & Warnings

  • Angioedema can occur and may be associated with life threatening respiratory compromise
  • Discontinue immediately in case of hypersensitivity reactions
  • Increase the risk of suicidal thoughts or behavior
  • Respiratory depression may occur when used with concomitant CNS depressants
  • May cause dizziness and somnolence
  • Withdraw gradually over a minimum of 1 week

Use in Special Populations

  • Safety and effectiveness not established for pediatric patients with certain conditions
  • Safety and effectiveness not established for Neugalin XR extended-release tablet in pediatric patients

Overdose Effects

  • Signs and symptoms include reduced consciousness, depression/anxiety, confusional state, agitation, restlessness, seizures, heart block

  • Primary anti-epileptic drugs
