Description
- An orally dissolving film designed to be applied on the tongue where it dissolves within 20 seconds and is then swallowed with saliva. It does not require water for dissolution or swallowing.
Indications
- Prevention of nausea and vomiting associated with emetogenic cancer chemotherapy
- Prevention and treatment of post-operative nausea and vomiting
- Prevention of radiotherapy-induced nausea and vomiting
Pharmacology
- Selective blocking agent of the serotonin 5-HT3 receptor type
- Potent and highly selective 5HT3 receptor-antagonist
- Blocks the initiation of vomiting reflex by activating vagal afferents via 5HT3 receptors
Dosage
- Dosage varies for different types of nausea and vomiting (chemotherapy-induced, radiotherapy-induced, postoperative)
- Different dosages specified for adults, pediatric patients, and different types of cancer chemotherapy
Administration
- Film dissolves on the tongue within 20 seconds and is then swallowed without water
- Hand should be washed after taking the film
Interaction
- Metabolized by hepatic cytochrome P-450 drug-metabolizing enzymes
- Clearance and half-life may be altered by inducers or inhibitors of these enzymes
Contraindications
- Hypersensitivity to the drug or any of its components
- Concomitant use of apomorphine
Side Effects
- Frequently reported adverse events: headache, constipation, diarrhea
- Rare cases of anaphylaxis, bronchospasm, tachycardia reported
Pregnancy & Lactation
- No evidence of impaired fertility or harm to the fetus in animal studies
Precautions & Warnings
- Hypersensitivity reactions reported in patients who have exhibited hypersensitivity to other 5-HT3 receptor antagonists
- Should not be used instead of nasogastric suction after abdominal surgery
Use in Special Populations
- Dosage adjustments not needed for patients with impaired renal function
- In patients with severe hepatic impairment, a lower recommended daily dose is advised
