Indications
- Short-term management of moderate to severe acute post-operative pain

Description
- Belongs to the pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits synthesis of prostaglandins
- Biological activity associated with the S-form
- Highly protein-bound
- Metabolized in the liver
- Metabolites and some unchanged drugs are excreted in urine
Pharmacology
- Potent analgesic
- Inhibits the cyclooxygenase enzyme system
- Minimal anti-inflammatory effect at its analgesic dose
Dosage & Administration
- Tablet: Recommended dose is 10 mg every 4-6 hours, short-term use only
- Injection: Used for the management of moderately severe, acute pain
- Analgesic effect begins within 30 minutes with maximum effect in 1 to 2 hours
- Different dosing for different age groups and conditions
- Eye Drops: 1 drop in each eye 4 times daily
Interaction
- Other NSAIDs or aspirin: Increase the side effects
- Anti-coagulants: Enhance anti-coagulant effect
- Beta Blocker: Reduce the anti-hypertensive effect
- ACE Inhibitors: Increase the risk of renal impairment
- Methotrexate: Enhance the toxicity of methotrexate
Contraindications
- Hypersensitivity to the drug or other NSAIDs
- Children under 16 years of age
- Prophylactic analgesic before surgery

- Eye Drops: Discontinue if corneal epithelial breakdown is present, avoid use with contact lenses
