Liptor EZ 20 mg+10 mg (Tablet)

Indications

• Adjunct to diet for atherosclerotic vascular disease
• Reduction of elevated total cholesterol
• Reduction of LDL-C in patients with homozygous familial hypercholesterolemia
• Increase in high-density lipoprotein cholesterol (HDL-C)
• Reduction of non-HDL-C
• Therapy for primary hyperlipidemia
• Therapy for mixed hyperlipidemia

Pharmacology

• Atorvastatin lowers plasma cholesterol and lipoprotein levels
• Inhibition of HMG-CoA reductase
• Reduction of LDL production
• Ezetimibe reduces blood cholesterol by inhibiting absorption
• Inhibition of cholesterol absorption by the small intestine
• Reduction of hepatic cholesterol stores
• Increased clearance of cholesterol from the blood

Dosage & Administration

• Daily dosage range of 10/10 mg to 80/20 mg
• Starting dose of 10/10 mg or 20/10 mg
• Administration as a single dose with or without food
• Dose adjustment based on lipid levels
• Specific dosage for patients with homozygous familial hypercholesterolemia
• No dosage adjustment necessary for patients with renal impairment

Interaction

• Increased risk of myopathy with fibric acid derivatives
• Coadministration with niacin and cyclosporine
• Avoid concurrent use with gemfibrozil
• Monitoring required when concomitantly used with digoxin
• AUC values consideration when selecting an oral contraceptive

Contraindications

• Active liver disease
• Unexplained persistent elevations of hepatic transaminase levels
• Hypersensitivity to any component

Side Effects

• Rhabdomyolysis
• Myopathy
• Liver enzyme abnormalities
• Myalgia
• Abdominal pain

Pregnancy & Lactation

• Contraindicated in pregnant women
• No benefit during pregnancy
• Unknown excretion in human milk

Precautions & Warnings

• Risk of myopathy with colchicine coadministration
• Temporary withholding in case of myopathy
• Liver enzyme tests prior to initiation
• Caution with concomitant drugs affecting endocrine function

Use in Special Populations

• Safety and effectiveness not established in pediatric patients
• No dosage adjustment necessary in geriatric patients

• Symptomatic and supportive measures
