Description
- Essential for calcium and phosphate absorption
- Prevents rickets, osteomalacia, and osteoporosis
- Aids in preventing pre-eclampsia during pregnancy

- Metabolized to 25-hydroxy Vitamin D3 (Calcidiol) in the liver

- Stored in tissues such as fat and muscle
- Eliminated via feces and urine
- Interacts with Magnesium containing Antacids, Digoxin, Thiazide Diuretics, Cholestyramine, Cholistipol, Phenytoin, Phenobarbital, Orlistat, Mineral Oil, and Corticosteroids
- Contraindicated in diseases associated with hypercalcemia
- Contraindicated in patients with known hypersensitivity to Vitamin D3
- Contraindicated in evidence of Vitamin D3 toxicity
- Symptoms of overdose include hypervitaminosis D

Pharmacology
- Binds to Vitamin D receptors (VDRs)
- Half-life of about 50 days as it is fat-soluble
- Requires measurement of 25 hydroxyvitamin D for maintenance therapy confirmation
- Increases calcium absorption
Dosage & Administration
- For adults: varying doses for treatment, prevention, and specific therapy for osteoporosis
- For children (12-18 years): varying doses for treatment and prevention
- Different dosage forms such as capsule, film-coated tablet, oroflash or chewable tablets, syrup, and injection

- Interacts with various drugs leading to deficiencies and increased need
Contraindications
- Contraindicated in diseases associated with hypercalcaemia

Precautions & Warnings
- Plasma-calcium concentration should be monitored at intervals
- Consultation required in renal impairment, pregnancy, and lactation
- Patients using Digoxin and Thiazide Diuretics should consult a health care practitioner before supplementing
