Indications
- Reduction of elevated total cholesterol
- Reduction of low density lipoprotein cholesterol
- Increase of high-density lipoprotein cholesterol
- Treatment of primary hyperlipidemia
- Treatment of homozygous familial hypercholesterolemia
- Adjunct to diet and nonpharmacologic measures
Pharmacology
- Inhibition of HMG-CoA reductase
- Cholesterol synthesis inhibition in the liver
- Increase in hepatic LDL receptors
- Reduction of LDL production
- Inhibition of absorption of cholesterol
- Localization at the brush border of the small intestine
Dosage & Administration
- Dosage range of 10/10 mg to 80/20 mg daily
- Recommended starting dose of 10/10 mg or 20/10 mg daily
- Single dose administration
- Analysis of lipid levels within 2 or more weeks
- Dosage adjustment according to lipid levels
- Dosage in patients with hepatic impairment
Interaction
- Increased risk of myopathy with fibric acid derivatives
- Enhanced risk of myopathy when combined with niacin
- Monitoring required when coadministered with digoxin
- Consideration of AUC values when coadministered with oral contraceptives
Contraindications
- Active liver disease
- Hypersensitivity to components of the preparation
