Indications

  • Reducing the risk of atherosclerotic vascular disease

Pharmacology

  • Atorvastatin lowers plasma cholesterol and lipoprotein levels
  • Ezetimibe reduces blood cholesterol by inhibiting absorption in the small intestine
  • Molecular target of ezetimibe is the sterol transporter NPC1L1

Dosage & Administration

  • Dosage range of 10/10 mg to 80/20 mg daily
  • Recommended starting dose of 10/10 mg or 20/10 mg daily
  • Dosage adjustment based on lipid levels
  • Specific dosage for patients with homozygous familial hypercholesterolemia
  • No dosage adjustment required for patients with renal impairment

Interactions

  • Increased risk of myopathy with concurrent use of fibric acid derivatives
  • Coadministration cautions with cyclosporine, niacin, and strong CYP3A4 inhibitors
  • Enhanced risk of skeletal muscle effects when combined with niacin
  • Monitoring required when coadministered with digoxin or warfarin

Contraindications

  • Active liver disease
  • Hypersensitivity to any component of the preparation
