Indications
- Treatment of adult patients with symptomatic anemia associated with chronic kidney disease (CKD)
Description
- Orally administered HIF-PH inhibitor promoting erythropoiesis through increasing endogenous production of erythropoietin, improving iron regulation, and overcoming EPO-suppressive effects of inflammation on hemoglobin synthesis
Pharmacology
- Orally bioavailable, HIF-PHI with potential anti-anemic activity, increases endogenous erythropoietin production, reduces hepcidin expression, improves iron availability, and boosts hemoglobin levels
Dosage & Administration
- Oral administration three times per week, individualized dose to maintain target Hb levels of 10 to 12 g/dL, starting dose based on erythropoiesis-stimulating agent treatment or patient weight, dose adjustments, method of administration with or without food
Interaction
- Potential drug-drug interactions with phosphate binders, gemfibrozil, probenecid, and other medications containing multivalent cations, recommendations for timing and dose adjustments
Side Effects
- Common adverse reactions including hypertension, vascular access thrombosis, diarrhea, peripheral edema, hyperkalemia, and nausea
Pregnancy & Lactation
- Contraindicated during pregnancy and breastfeeding
Precautions & Warnings
- Caution in patients with pre-existing risk factors for thrombotic vascular events, history of seizures, signs of infection, or liver disorder, contraindicated in pregnant women
Use in Special Populations
- Not indicated in children, not recommended for use in patients with severe hepatic impairment
Overdose Effects
- Symptoms of increased heart rate transient and likely excessive hemoglobin concentration, treatment with appropriate measures of dose reduction or interruption, not removed by dialysis
