Formulation

Dosage & Administration
- Adults: 40000 IU once weekly for 7 weeks for Vitamin D3 deficiency
- Children (12-18 years): 20000 IU once every 2 weeks for 6 weeks for Vitamin D3 deficiency
- Adults: 1000 IU (1-2 tablets) daily for film-coated tablet
- Children: 1000-2000 IU daily for oroflash or chewable tablets
- 0-1 yr: 400 IU/ day (2 ml) for syrup
- Vitamin D enriched milk: ½ ampoule (0.5 ml) i.e. 1,00000 I.U. every 6 months for injection
Mode of Action
- Calcitriol binds to Vitamin D receptors for calcium and phosphate absorption
- Vitamin D3 has a half life of about 50 days
- Absorbed in the small intestine and metabolised to 25-hydroxy Vitamin D3
- Second hydroxylation to 1,25-dihydroxy Vitamin D3 occurs in the kidney
Interaction
- Interferes with absorption and metabolism of Vitamin D3: Magnesium containing Antacids, Digoxin, Thiazide Diuretics, etc.
Contraindications
- Hypercalcaemia
- Known hypersensitivity to Vitamin D3
- Evidence of Vitamin D3 toxicity

- Nausea & Vomiting
- Weight Loss
- Headache
- Raised concentrations of calcium and phosphate in plasma and urine
Pregnancy & Lactation
- Safe use of doses up to 4000 IU during pregnancy
- Excreted in breast milk
- Recommended daily intake for pregnant women is 400 IU
Precautions & Warnings
- Monitor plasma-calcium concentration during high doses
- Consult health care practitioner when using Digoxin and Thiazide Diuretics
- Consult health care practitioner in liver or kidney disease, primary hyperthyroidism, etc.
Use in Special Populations
- Safety & efficacy in children under 12 years not established
