Indications
- First-line treatment for metastatic non-small cell lung cancer (NSCLC)
- Indicated for patients with EGFR exon 19 deletions or exon 21 L858R mutations
- Treatment for metastatic EGFR T790M mutation-positive NSCLC
Mode of Action
- Kinase inhibitor of epidermal growth factor receptor (EGFR)
- Binds irreversibly to mutant forms of EGFR
- Inhibits activity of HER2, HER3, HER4, ACK1, and BLK
Dosage & Administration
- Recommended dose of 80mg tablet once daily
- Can be taken with or without food
- Disperse tablet in non-carbonated water only
- Do not crush, heat, or ultrasonicate
Interactions
- Avoid concomitant administration with strong CYP3A inhibitors
- Effect on drugs sensitive to CYP3A, BCRP, or CYP1A2 with narrow therapeutic indices
Side Effects
- Common adverse reactions include diarrhea, rash, dry skin, nail toxicity, stomatitis, fatigue, decreased appetite
Pregnancy & Lactation
- Can cause fetal harm in pregnant women
- Advise effective contraception for females and male partners of reproductive potential
- May impair fertility in females and males of reproductive potential
Precautions & Warnings
- Risk of Interstitial Lung Disease (ILD)/Pneumonitis
- Monitor for QTc Interval Prolongation
- Assess left ventricular ejection fraction (LVEF) before treatment
Use in Special Populations
- No clinically significant differences based on age, sex, ethnicity, renal or hepatic impairment
- Safety and effectiveness not established in pediatric patients
- No recommended dose for patients with severe renal or hepatic impairment
