Indications
- Treatment of Mantle Cell Lymphoma (MCL)
- Treatment of Chronic Lymphocytic Leukemia/Small Lymphocytic Lymphoma (CLL/SLL)
- Treatment of Waldenstrom Macroglobulinemia (WM)
- Treatment of Marginal Zone Lymphoma (MZL)
- Patients who have received at least one prior therapy
Pharmacology
- BTK inhibitor
- Forms a covalent bond with a cysteine residue
- Inhibits BTK enzymatic activity
- Inhibits malignant B-cell proliferation
- Affects B-cell trafficking, chemotaxis, and adhesion
Dosage
- Recommended dose for MCL and MZL: 560 mg once daily
- Recommended dose for CLL/SLL and WM: 420 mg once daily
- Combination dosage with bendamustine and rituximab
Administration
- Orally administered
- Take once daily at the same time
- Swallow capsules whole with water
Interaction
- Avoid co-administration with strong and moderate CYP3A inhibitors
- Dose adjustment when used with moderate CYP3A inhibitors
- Avoid co-administration with strong CYP3A inducers
Contraindications
- Hypersensitivity to the active substance or excipients

- No overall differences in effectiveness in geriatric patients
- Increased frequency of anemia and pneumonia in older patients
- Metabolized in the liver
- Safety not evaluated in patients with hepatic impairment
Overdose Effects
- No specific experience in the management of overdose
- Reversible Grade 4 hepatic enzyme increases observed in one case
- Closely monitor and provide appropriate supportive treatment
