Indications

  • Invasive aspergillosis
  • Candidemia
  • Disseminated candidiasis
  • Esophageal candidiasis
  • Serious infections caused by Scedosporium apiospermum
  • Refractory therapy

Pharmacology

Inhibits ergosterol synthesis by inhibiting CYP450-dependent 14-alpha sterol demethylase

Dosage & Administration

  • Oral:
    • To be taken at least one hour before or after a meal
  • Injection:
    • Invasive Aspergillosis:
      • Loading dose: 6 mg/kg 12 hourly for the first 24 hours
      • Maintenance Dose: 4 mg/kg 12 hourly
    • Candidemia:
      • Loading dose: 6 mg/kg 12 hourly for the first 24 hours
      • Maintenance Dose: 3-4 mg/kg 12 hourly
    • Scedosporiosis and Fusariosis:
      • Loading dose: 6 mg/kg 12 hourly for the first 24 hours
      • Maintenance Dose: 4 mg/kg 12 hourly

Interaction

  • Adjust Viera dosage and monitor for adverse reactions
  • May increase the concentrations and activity of drugs that are CYP3A4, CYP2C9 and CYP2C19 substrates
  • Increase maintenance oral and intravenous dosage with co-administration of certain drugs

Contraindications

  • Known hypersensitivity to Voriconazole or any other components of this drug
  • Co-administration with specific drugs due to risk of serious adverse reactions or loss of efficacy
