Indications
- Relief of signs and symptoms of osteoarthritis
- Relief of signs and symptoms of rheumatoid arthritis
Pharmacology
- Exhibits anti-inflammatory properties
- Exhibits analgesic properties
- Exhibits antipyretic properties
- Prodrug with hepatic biotransformation to active component 6-methoxy-2-naphthylacetic acid (6MNA)
Dosage & Administration
- Dosage: 500mg & 750mg tablet
- Dosage: 1000mg tablet mixed in a glass of water
- Recommended starting dose: 1000mg as a single dose with or without food
- Maximum starting doses for patients with renal insufficiency: 750mg or 500mg once daily
- Use in pediatric patients not established
Interaction
- NSAIDs may diminish the antihypertensive effect of ACE-inhibitors
Contraindications
- Contraindicated in patients with known hypersensitivity to nabumetone or its excipients
- Contraindicated in patients who have experienced asthma, urticaria, or allergic reactions after taking aspirin or other NSAIDs
- Contraindicated for peri-operative pain in the setting of coronary artery bypass graft (CABG) surgery
Side Effects
- Gastrointestinal: Diarrhea, dyspepsia, abdominal pain, constipation, flatulence, nausea, positive stool guaiac, dry mouth, gastritis, stomatitis, vomiting
- Central Nervous System: Dizziness, headache, fatigue, increased sweating, insomnia, nervousness, somnolence
- Dermatologic: Pruritus, rash
- Special Senses: Tinnitus
- Miscellaneous: Edema
Pregnancy & Lactation
- Pregnancy Category C
- Use during pregnancy only if needed
- Use during the third trimester not recommended
- Not recommended for use in nursing mothers
Precautions & Warnings
- Associated with renal papillary necrosis and abnormal renal pathology in animals
- Associated with reduction in renal blood flow in patients with conditions leading to a reduction in renal blood flow or volume
- Those at greatest risk: patients with impaired renal function, heart failure, liver dysfunction, those taking diuretics, and the elderly
- Discontinuation may lead to recovery to pretreatment state
Overdose Effects
- Symptoms following acute NSAIDs overdoses: lethargy, drowsiness, nausea, vomiting, epigastric pain
- Rare effects: hypertension, acute renal failure, respiratory depression, coma
- Anaphylactoid reactions reported with therapeutic ingestion of NSAIDs
- Management: symptomatic and supportive care, no specific antidotes
- Reported overdoses of up to 25g with no long-term sequelae following standard emergency treatment
