Indications
- Treatment of iron overload patients with thalassemia major
- Alternative when deferoxamine therapy is contraindicated or inadequate
- Initiated and maintained by experienced physician
Pharmacology
- Chelating agent with affinity for ferric ion
- Forms stable 3:1 complexes with ferric ions
- Lower binding affinity for other metals like copper, aluminum, and zinc
Dosage & Administration
- Oral administration for adults and children over 6 years
- Doses of 25mg/kg three times daily
- Maximum recommended daily dose of 100mg/kg
Interaction
- Caution with concurrent use of Defera and Vitamin C
- Avoid concurrent use with aluminium-containing antacids
Contraindications
- Hypersensitivity to the active substance or any excipient
Side Effects
- Common gastrointestinal disorders like diarrhea, nausea, vomiting, and abdominal pain
- Reddish brown discoloration of urine
- Reported adverse effects include arthralgia and increased liver enzymes
Pregnancy & Lactation
- Not recommended for use in pregnant and lactating women
Precautions & Warnings
- Causes neutropenia including agranulocytosis
- Neutrophil count monitoring required
- Caution advised in patients with hepatic or renal impairment
