Indications
- Treatment of gestational choriocarcinoma, chorioadenoma destruens and hydatidiform mole

Pharmacology
- Inhibition of dihydrofolic acid reductase
- Interference with DNA synthesis and cellular replication
Dosage
- Oral or intramuscular administration
- Different doses for different diseases
- Doses for neoplastic diseases, acute lymphoblastic leukemia, lymphoma, mycosis fungoides, breast cancer, head and neck cancer, rheumatoid arthritis, polyarticular-course juvenile rheumatoid arthritis, and psoriasis
Administration
- Best taken on an empty stomach
- May be taken with meals to reduce GI discomfort
- Avoid taking with milk-rich products
Interaction
- Interactions with NSAIDs, salicylates, phenylbutazone, phenytoin, sulfonamides, penicillin, theophylline, probenecid, azathioprine, retinoids, sulfasalazine, cisplatin, mercaptopurine, tetracycline, chloramphenicol, nonabsorbable broad spectrum antibiotics, vitamin preparations containing folic acid or its derivatives, and trimethoprim/sulfamethoxazole
Contraindications
- Alcoholism or other chronic liver disease
- Immunodeficiency syndromes
- Pre-existing blood dyscrasias
- Hypersensitivity to methotrexate
- Pregnancy, lactation, and women of childbearing potential
